What is the source of this du’a?
Allahummahdina wahdi bina wa yassiril huda lana, waj ‘alna mafatiha lil khayri wa maghalika lish sharr
(O Allah! guide us and guide others through us, make guidance easy for us, and make us the source of good and a barrier/locks against evil)

I have not come across this as one du’a in a Hadith. Nonetheless, there is substantiation for this recital. See below for details:
1) The words ‘Allahummahdina wahdi bina’ appear in a narration recorded in Musannaf ‘Abdur Razzaq. See here.
2) Imam Tirmidhi and other Muhaddithun have recorded a du’a of Nabi (sallallahu ‘alayhi wa sallam), wherein Nabi (sallallahu ‘alayhi wa sallam) would say, ‘Wahdini wa yassiril huda li’ (Guide me and make guidance easy for me). (Sunan Tirmidhi, Hadith: 3551)
3) The words ‘Waj ‘alna mafatiha lil khayri wa maghalika lish sharr’ seem to be formed from the Hadith recorded in Sunan Ibn Majah. see the full Hadith here.
There is no harm in reciting the du’a in your question, as it is based on a few Hadiths.
