If ghusl becomes obligatory and she takes ghusl from a bucket of water, will the water become mustamal even if hands are washed before putting it into the bucket? I have read that the water becomes mustamal only when the hands are not washed before putting it into the bucket.

No, this water does not turn used up (musta’mal), only the water which is used with the intention of wudhu or ghusl or removal of hadath (impurity) becomes musta’mal.
